The authors report studies on kinetic characteristics of thrombin receptor-mediated responses, i.e., Ca2+- activated K+ currents, in rat megakaryocytes, using a rapid drug exchange system. Overall, the thrombin responses were compared with similar responses induced by activation of purinoceptors with ATP. It is shown that, in comparison to the ATP response, the thrombin-induced response is characterized by a long latency, a long washout time and a fast desensitization. In addition to thrombin, the megakaryocyte responses to trypsin and a synthetic thrombin receptor agonist peptide, were also studied. In general, this is a well performed study containing new interesting data which deserves publication.

Referee's Comments

 

It is shown in this paper that thrombin activates a Ca2'-dependent K+ current in megakaryocytes. Although it is not proven beyond doubt that the current is an IKCa it probably is.

 

The phannacological results are convincing and the paper deserves publication. However, the authors need to corrcct frequent language errors. I have picked up some errors that are listed below but have probably missed many others.
